چکیده

Five to ten percent of HIV-1 infected individuals remain asymptomatic up to a year after infection. This type of infection is known as a long-term non-progressive (LTNP) infection, and can be characterized as a viral phenotype with a lack of CD4+ Tcell death. This MQP deals specifically with a group of LTNPs known as the French ALT group who have a form of the virus that is considerably less cytopathic. Because mutations in the Viral Protein R (Vpr) region of the HIV-1 genome have been observed to cause similar viral phenotypes, the ALT patient Vpr alleles were investigated. Several Vpr polymorphisms were identified in hypervariable loci, most of which result in significant amino acid changes. In the future, these mutations will be mimiced in wildtype HIV’s to determine their effect on viral replication to determine if they are responsible for the LTNP phenotype.

Temporal relationship between V1V2 variation, macrophage replication, and coreceptor adaptation during HIV-1 disease progression.

BACKGROUND Specific mutations in VPR and V2 potentially restrict HIV-1 replication in macrophages. Such restriction could potentially limit HIV replication in long-term non-progressors (LTNP), thus accounting for low viral load and delayed progression to AIDS. OBJECTIVE To examine whether a specific VPR phenotype (truncated versus non-truncated) correlates with disease progression and whether...

Evidence for a cytopathogenicity determinant in HIV-1 Vpr.

HIV-1 is cytopathic for CD4(+) T lymphocytes in vitro and this property of HIV-1 is generally considered to account for some of its in vivo cytopathogenicity. Thus, the extent of lymphocyte depletion correlates with the level of viremia whereas low levels of viral replication are typically associated with stable lymphocyte levels and asymptomatic infection such as is observed in non-progressors...

Characterization of the Molecular Determinants of Primary HIV-1 Vpr Proteins: Impact of the Q65R and R77Q Substitutions on Vpr Functions

Although HIV-1 Vpr displays several functions in vitro, limited information exists concerning their relevance during infection. Here, we characterized Vpr variants isolated from a rapid and a long-term non-progressor (LTNP). Interestingly, vpr alleles isolated from longitudinal samples of the LTNP revealed a dominant sequence that subsequently led to diversity similar to that observed in the pr...

Antibodies to Tat and Vpr in the GRIV cohort: differential association with maintenance of long-term non-progression status in HIV-1 infection.

The HIV-1 regulatory protein Tat and the accessory protein Vpr are thought to stimulate viral replication and contribute to viral pathogenesis as extracellular proteins. Humoral immune responses to these early viral proteins may therefore be beneficial. We examined serum anti-Tat and anti-Vpr IgG by ELISA in the GRIV cohort of HIV-1 seropositive slow/non-progressors (NP) and fast-progressors (F...

Differences in disease progression in a cohort of long-term non-progressors after more than 16 years of HIV-1 infection.

BACKGROUND It is unclear whether resistance to immunologic damage in long-term non-progressors (LTNP) will last indefinitely or whether it merely represents the extreme of a Gaussian distribution, and therefore progression will occur eventually. PATIENTS AND METHODS A cohort of 19 LTNP was established in 1997. Plasma viraemia and CD4 cell counts were measured two to three times each year unti...
